Process DOSY data with T1T2 Application in Topspin:

  1. In topspin, load dosy “2D” data, rser 1 ↙, efp↙, apk↙, absn↙ .ph, nD save (important!), and save & exit,
  2. load dosy “2D” data again, xf2↙, abs2↙ (may try absg = 1 or 5 and see different effect, — degree of polynomial for abs, [0..5] (F2,F1))
  3. setdiffparm↙
  4. Applications → Dynamics → T1T2 → FID →Spectrum → 1
  5. Peaks/Ranges → Manual integration → define Ranges → integrate regions (choose more peaks…) → export to relaxation module.
  6. Go to Relaxation tab → check parameters (make sure to choose: 1000, -1000 for baseline correction limits; vargrad, difflist, etc.) → press “>” to fitting, click full spectrum button, click sq, and then click lg; and then press “>>>” to fit all other points, then click “show report”, save into a text file.

Note:

  • d20 (e.g. 30ms here) *2 = big Delta (∆),
  • P30 (e.g. 1ms here) *2 = little Delta (d)  (for pulse sequence using ste bipolar gradients, d = p30*2; for one using spin echo gradients, d = p30; also look at the pulse sequence graphic, previous one has 4 gradients; later one has two gradients), software picks them up automatically by macro “setdiffparm” already.

  1. In topspin, load dosy “2D” data, edp↙, rser 1 ↙, efp↙, apk↙, absn↙ .ph, nD save (important), and save & exit.
  2. load dosy “2D” data again, xf2↙, abs2↙
  3. setdiffparm↙
  4. eddosy↙ (will open dosy processing panel, make sure: lb=2Hz; absg (degree of polynomial for abs) =1; LWF (line width factor) = 5; PC=10, noise sensitivity factor; DISPmin: -10; DISPmax: -8; choose “intensity”)
  5. dosy2d setup (automatically preset some parameters, won’t alter the data)
  6. dosy2d
